Methotrexate is dosed at 0.6–0.8 mg/kg IV Per protocol in dogs, per USPC. Veterinary Information- Appendix III. 1990;2:2811-2860. — Plumb's 10th ed printed p.856, Methotrexate monograph spanning p.856-858. Verify against current literature before clinical use.
Inhibits dihydrofolate reductase, blocking thymidylate and purine synthesis. Impairs DNA replication in rapidly dividing cells.
Myelosuppression, GI toxicity (mucositis, diarrhea), hepatotoxicity, nephrotoxicity. Leucovorin rescue may be needed. Not commonly used as single agent in veterinary oncology. Monitor CBC and renal function.
